Hello , I uncovered my boat the other day getting ready for the season. Put the muffs on motor started right up, but no water being sucked up and out exhaust, I changed impeller with a new impeller kit, new housing, seals everything was included. Put outdrive in a barrel instead of the muffs so drive is completely submerged, still no water !! I changed water ( circulation) pump, still no water !! I made sure the water tube didn’t fall off when putting back together. 4.3 mercruiser alpha 1 gen 2

Did you check the water inlet screens? are they clogged? Did you ream out your 'pee' hole? Could be that an insect has made its home in the pee tube and it's full of bug nest dirt. It has been known to happen.

Screens are clean, but I did not clean pee hole. I did unscrew blue screw on bottom of manifold and no water came out.

I don't know much about an I/O V-6. Didn't know they had a pee hole. I'm guessing the V-6 is similar in design as a car engine and if the thermostat is stuck in the closed position, it wouldn't let water to circulate through the engine. I'd check or change the thermostat since you've already changed the water pump.

I didn't realize that water sucked in through the outdrive? I always thought the water pickup was thru the hull..

I took off hose coming from outdrive to thermostat housing, put garden hose in and water came out the outdrive intake holes with no problem !!

Yep, still sounds like the thermostat is closed. I'd take the thermostat out and then see if water comes out the outdrive. If it does then replace the thermostat.
BTW: I'm assuming you are running it long enough for the engine to heat up and the thermostat open as it should. You should be able to hold your hand on the engine block. It should warm up but not get so hot that you can't keep your hand on it. If it gets too hot to keep your hand on the block, the thermostat isn't opening as it should and no cooling water is circulating through the engine.

I replaced thermostat with new Tstat kit , only issue was the sleeve that holds down thermostat had to ordered separately, I ran garden hose through all hoses coming from Tstat housing and the water came out as it should, put it all back together, she started right up and water is coming out as it should !!! Thanks again for all your help !! ......its funny all the mercruiser dealers around my way and mercruiser parts stores said the thermostat has nothing to with it. If you (google) look at the water flowdiagram of the alpha 1 gen 2 it looks like thermostat has nothing to do with it either !!

Here we go again !!!! Put muffs on, started her up, no water !!! Took hoses off tstat housing put garden hose in and water came out as it should, tightened hoses back on tstat, muffs on good pressure on the muffs ,started her up and water is coming out again !!! I ordered new water pick up tube and gasket, (original one is copper or brass, new one is plastic) that was a little beat up when I did the impeller ,I think it's getting air somewhere in the outdrive because when the manifold and outdrive are filled with water, it works correctly so I think the water I am putting in is almost like priming it , make sense ?

Theres a check valve on there that allows the water to bypass the tstat while its closed, also allows water to come into the flush port. When the system is not clean they get stuck and when they wear a bit they get stuck too.
Im not familiar with your motor so I cannot tell you exactly where it is. You have to look it up but check there for sure.

When I took outdrive off again !! I noticed that the water hole on upper end of outdrive was egg shaped and not round (looked worn out ) so would not match the hole on gimbal housing and would not make a tight seal, causing it to suck up air and not water !! (I think) I'm trying to figure out how to send you guys a picture.
